My idea: 1. rinse under tab water with liquid soap. 2. 15min in 10% bleach 3. 3times rinse in steril water Should i dip in 70% Iso? Marc S. Posted October 20, 2014 Report Share Posted October 20, 2014 Hi Tobias, I would add some detergent (0.5% Triton-X) - that breaks the surface tention! You could try to wash them first with water and then sterilze them with two EtOH concentrations (1st: 100%, 2nd: 50%) with detergent and a last "wash" with pure (or maybe better 70%) EtOH (without detergent)... It calls simple! @Bux: you mean 15min in 10% bleach is sufficient? You mean the contaminants are inside the roots? I think surface sterilisation is sufficient! You don't think so? I have tried some: 1. week good 2. week good The third and fourth week is ok too, without contamination In the fifth week i have a nice mushroom culture.....Sterilisation with ppm solution for 6h, 3x rinse for 5min each. But i don't give up ;0) Could i try the roots with a similar steri protocol? PPM solution could work good...
