I want use smoke water to pretreatment, question is when, before sterilization or after that but then seed will be probably contaminated . Is there anyone who have experience with treatment GA3 or with smoke water then move seed to invitro process :?: I hope there are some people who try invitro of hard germinated seed like byblis, tuberous drosera ... Easier than germinating seed if so. I successfully placed U. longifolia in vitro from leaf pieces. These were just the usual 1% sodium hypochlorite for 10mins and 5x water washes. The leaf was sterilised whole and then cut before placing on 1/3 MS salts & vitamins + 20g/L sucrose + 6g/L agar + 1.0mg/L BAP. Plantlets formed from leaf and petiole tissue. Raging success.
